The sample to be examined is planted in a suitable agar medium in a petri dish using the single colony drop technique or the spreading technique. Petri dishes are incubated at the appropriate temperature and time. After incubation, the colony formed in petri dishes, not in contact with each other and suitable for the characteristics of the microorganism to be isolated, is transferred to a liquid medium with the help of a loop, toothpick or grafting needle. After incubation, purity control is performed to examine whether the isolate is pure.
For purity control, the agar medium in the petri dish is smeared and the characteristics such as shape, size, structure and color of the colony that will form here after incubation are examined. When the presence of colonies with different characteristics is observed, this means that the culture is mixed and the procedures are repeated until the purity control by taking a single colony again. Even if the same type of colony is observed in the medium, it is generally recommended to perform reisolation by taking a single colony.
